7 Alyx System Draw Phase Quality, speed and simplicity in double red cell collection Approximately 300 mL of whole blood is withdrawn form the donor during each draw phase. ACD-A is added immediately to whole blood at a ration of 1:11. The anticoagulated whole blood is simultaneously pumped into the in-process container and the separation chamber. The centrifuge separates the whole blood into plasma and red cells. Plasma is drawn into the plasma container and red cells into the red cell container. Only the amount needed to reach the target red cell volume is drawn during the final draw phase.

8 Alyx System Return Phase
Quality, speed and simplicity in double red cell collection During each return phase, plasma and saline are returned to the donor, while the centrifuge continues to separate whole blood. This continuous processing contributes significantly to the greatly reduced procedure time. Donor comfort and safety are also enhanced as the donor remains relatively isovolemic throughout the procedure. Because the saline and plasma are mixed together before they are returned to the donor, they are closer to body temperature and the return is therefore more comfortable for the donor. In addition, several cycles of draw and return make adverse reactions far less likely than may be seen with other automated systems. Following each return phase, the next draw phase is automatically initiated. This repeats for four to five cycles of draw and return until the target red cell volume is reached. During the final return phase, all excess blood components are returned to the donor along with saline, leaving the donor isovolemic.

9 Alyx System Leukoreduction Phase
Quality, speed and simplicity in double red cell collection Upon completion of the final return, the ALYX System automatically initiates the addition of SAG-M red cell preservation solution and the beginning of leukodepletion filtration. Red cell preservation solution is added to concentrated red cells at a ratio of 1 mL preservation solution per 2.1 mL absolute red cells. Red cells with preservation solution are pumped through a single leukodepletion filter and separated into two red cell storage containers The filtration process is performed without operator intervention and concurrently with postprocedure donor care.

Manual collection 방법 Collection Component Processing Leukoreduction Summary of Key Messages: In manual collection systems there is at least five steps in the entire process. Step one is to screen the donor; step two is to collect; step three is to process the unit (i.e. separate into components); step four is to leukoreduce and step five is to label the unit. ALYX is designed to integrate the collection, processing, and leukoreduction steps into one step. After collection, the units must be labeled and tested before they are available for distribution. 12 대상 수혈용 혈액으로 국내 처음으로 시도되는 것으로 국내 혈액관리법에 두 단위 적혈구 헌혈자 기준이 제정되어 있지 않아 미국FDA기준과 유럽 council의 기준에 준하여 선정 2008년2월~6월 건강한 성인 지원자 중에서 신장 170 cm 이상, 체중 70 kg이상, 혈색소 14.5 g/dl 이상인 남자 17명 헌혈유예기간 : 16주 (4개월) 모든 대상자는 헌혈 전 문진과 과거력 조사를 통하여 헌혈 적합 여부를 확인함

13 결과 헌혈자의 헌혈 전후 Hct ,Plt, WBC count의 변화를 Advia 2120을 이용하여 종료후 최대 30min 이내에 측정함. 최종 성분채집적혈구의 잔존 WBC count는 Nageotte chamber를 이용하여 측정함
