5 Confocal imaging – 개선사항 없음
P-chip Confocal imaging – 개선사항 없음 immunostaining은 cell과 target protein에 따라 conditioning을 다르게 해야 함 현재까지 JEG-3의 zo-1 염색한 논문은 하나밖에 없는데, 해당 논문의 방법대로 염색하면 위와 같이 제대로 안 나옴.. 아산병원 공초점현미경실 선생님 도움받아 진행 중

6 Cell detachment 해결(?) P-chip Fibronectin coating protocol 대폭 변경
DPBS washing  40ul/ml FN injection within 24hr after plasma treatment  Overnight incubation  Media injection  At least 2hr incubation  Cell seeding (counting) Seeding density & waiting time 에 따라 실험 구성 Chip # Seeding density Waiting time ① 6x107 cells/ml 2hr ② 1.2x107 cells/ml ③ 4hr ④ Results Highest success rate since May But Cell stock이 바뀌었으므로 완전히 FN coating 덕분인지는 알 수 없음… ① ② ③ ④

7 인슐린 교환 실험 – 구성 P-chip Normal human blood insulin level : 5~25 μU/ml
BUT in vitro insulin perfusion 실험 논문이 있으므로 이에 맞춰 진행할 예정 (아래 2가지 선택지 중 상의 후 결정) ① 100 or 200 농도만 ② 100, 1000 두 가지 농도
