General comments: 본인의 의견 추가 가능 Reviewer 1 This study investigated autophagy and NET production levels by neutrophils isolated from peripheral blood samples taken from patients with severe asthma or with less severe disease. The effects of NET on in vitro inflammatory functions were also examined in human airway epithelial cells or peripheral blood eosinophils. Overall this is a potentially interesting study but I have a number of queries. 1. My main concern is that from Table 1 30 SA patients and 38 NSA patients were recruited yet in Figure 1B data from 24 and 22 patients and for 1D 14 and 8 respectively are presented. The same issue applies to Figure 2 and 3. Was this simply due to lack of cells or was any selection made. If the latter is the case how was this done? Thank you for your comment. We described the study cohort in the “Materials and Methods” section (Lines 79-85) and “Results” section (Lines 223 & 232)., We recruited totally 68 asthmatic patients into 2 independent cohorts of with SA (n=30) and NSA (n =38) patients. In the first cohort, we investigated autophagy levels in PBNs isolated from 22 SA and 24 NSA patients using immunoblots. Because of limitations in number of PBNs that could be obtained from each study subject, we further recruited 8 SA and 14 NSA patients to the second cohort to evaluate neutrophil autophagy and NET production levels using confocal microscopy and PicoGreen assay.
