2 실험 배경 EZH2는 Histone H3 K27 methyltransferase 타겟 유전자의 발현을 억제시켜 lineage-specific한 proliferation과 differentiation에 관여함 T-cells, hematopoietic stem cells, neurons 등 다양한 세포에서의 기능은 밝혀져 있으나 mammary gland에서의 기능 연구는 부족함 실험 전략 및 방법 Development 동안 다양한 time points에서 mammary gland tissue 확보 (Virgin, p6, p13, L1) Immunofluorescence를 통해 mammary gland 내 EZH2의 localization 확인 Western blot을 통해 EZH2의 protein 양을 분석

3 실험 결과 1 Virgin p6 p13 L1 EZH2 B-actin Western blotting을 통한 EZH2 발현양 확인
Virgin, pregnant day 6 (p6), p13, lactation day 1 (L1)의 mammary gland tissue로부터 protein을 확보 20ug의 protein을 SDS-PAGE gel에 loading함 EZH2 antibody (1:1000) 과 loading control을 위해 B-actin (1:10000)사용함 EZH2의 발현은 virgin과 비교했을 때 pregnancy와 lactation 기간에 증가함을 확인

4 실험 결과 2 (B) Immunofluorescence를 통한 EZH2 localization 확인
Virgin, p6, p13, L1 mammary tissue를 formaldehyde에 고정하여 만들어진 paraffin block으로부터 section 진행 EZH2 (red, 1:100), E-cadherin (green, 1:200), DAPI (blue, 1:10000) EZH2의 발현이 pregnancy 기간 중 alveolar cell에서 강하게 발현됨을 확인

5 결론 EZH2는 pregnancy 기간에 양이 증가하기 시작하여 mid-pregnancy (p13)일 때 가장 높은 발현양을 보임 특히 Pregnancy 기간 중 proliferation과 differentiation이 일어나는 alveolar cell에서 높게 발현되는 것을 확인 따라서 EZH2가 alveolar cell의 proliferation과 differentiation에 관여하는 유전자들을 조절할 것으로 예상 EZH2의 타겟 유전자들과 기능을 검증하기 위해서 EZH2 유전자가 knock out 된 마우스를 제작하여 검증하는 것이 필요하다고 생각됨 참고문헌 Hennighausen L., Robinson G.W.
